TODAY we are going to talk about pathophysiology


The homogentisate 1,2-dioxygenase(HGD) requires 6 subunits to form 2 HGD trimer which in turn forms a bioactive HGD hexamer. The HGD uses Fe2+ and O2 to cleave the aromatic ring of the homogentisic acid(HGA)to form maleylacetoacetic acid.However, a mutation in one or more of the 6 subunits will interfere with the formation of the HGD trimer.Thus, there will not be any bioactive homogentisate 1,2-dioxygenase.

  1. Erm sry if Im a bit slow to understand this but, what would happen if there is no bioactive homogentisate 1,2-dioxygenase??

  2. If there is no bioactive homogentisate 1,2 dioxygenase, the Homogentisic acid(HGA) cannot be broken down to maleylacetoacetic acid. This will cause HGA build up in the body and HGA happens to be toxic. HGA oxidation to benzoquinone acid(BQA) in the urine will cause black urine and cause other complications such as ochronosis and osteoarthritis. U can read the previous blogs to have a more in depth understanding on the complications.
